DNA: the instruction book inside youDNA:

Almost every cell in your body carries a full copy of the instructions for building you.

3 billionletters in one full set of human DNA个字母,组成一套完整的人类DNA
~2 mof DNA folded up inside a single cell长的DNA,折叠在一个细胞里

A twisted ladder

DNA is a very long molecule shaped like a spiral ladder, called a double helix. The two rails hold it together. The rungs carry the message.DNA

Written with just 4 letters

A always pairs with T, and C always pairs with G. The order of the letters is the instruction, like words in a sentence.A总是和T配对,C总是和G配对。

02

Chromosomes and genes: how DNA is organized染色体与基因:DNA是怎样组织起来的

DNA is packed into chromosomes. A gene is one stretch of DNA that holds one instruction.DNA被打包成染色体。

~20,000genes in humans

Cell nucleus

The control room of the cell. It keeps your DNA safe — one very long DNA thread, coiled up tightly.

Genes as recipes

One stretch of DNA = one recipe. Humans have about 20,000 genes.

In each pair, one chromosome comes from the mother and one from the father. Pair 23 sets sex: XX typically for female, XY typically for male.

03

From gene to protein: reading the recipe

A gene is a recipe. A protein is the finished dish, and proteins do almost every job in your body.

In the nucleus, the gene is copied into a message called mRNA.

A ribosome reads the message and links amino acids into a chain.

3

3 · Fold into a worker3 · 折叠成工人

The chain folds up. Now it is a working protein.

Hemoglobin

Carries oxygen in your blood

Keratin

Builds your hair and nails

Enzymes酶

Break down the food you eat

Antibodies抗体

Catch germs that get in抓住入侵的病菌

04

Cells build tissues, organs and you细胞组成组织、器官,最终组成你

Cells team up into tissues, tissues into organs, and your organs keep you alive every second.

30+ trillioncells in an adult body

Digestive system

9 meters of tube that turns food into fuel

Why these three matter first为什么先看这三个

If the brain or heart stops, cells start to die within minutes. Without digestion, every cell runs out of fuel.

05

One genome, many kinds of cell

Every cell carries the same DNA. What makes a cell a nerve cell or a muscle cell is which genes it switches on.

~20,000protein-coding genes found so far, plus 35,000+ RNA-only genes个蛋白质编码基因,另有3.5万多个只产生RNA的基因
1 in 5is well studied; what the other ~16,000 do is still largely unknown
4,213genes tied to a single-gene disease so far — and the list keeps growing

Young red blood cell

Turns on the hemoglobin gene → makes hemoglobin → carries oxygen

06

How parents pass their genes to a baby

Each parent passes on a random half of their chromosomes. Together they make one complete new set.

1

A random half from each parent

For each of the 23 pairs, chance decides which chromosome goes into an egg or sperm — like a coin flip.

2

Together: a full set

They join into one cell with 46 chromosomes. That cell divides again and again to build the baby.

3

A new mix every time每次都是新组合

Each parent can make over 8 million combinations, so brothers and sisters are alike but not the same.

07

How many diseases are gene-related?

Nearly all diseases involve genes, but only a few thousand are caused by genes alone. Sorted by how much genes matter, diseases fall into three groups.

Genes are the cause · High

72% of the 6,000+ known rare diseases are genetic. About 300 million people (≈4 in 100) live with a rare disease. One faulty gene or chromosome is usually enough. Examples: sickle cell disease, cystic fibrosis, Down syndrome.

Genes shift the risk · Medium

About 40% of 560 common conditions show a clear inherited part. 3 in 4 deaths worldwide come from this group. Many genes each add a little; lifestyle and environment add the rest. Examples: heart disease, type 2 diabetes, most cancers.

Outside causes lead · Low

1 in 4 deaths worldwide come from this group. A germ, an injury, or a lack of food is the trigger. Genes still affect how hard it hits you. Examples: flu, tuberculosis, road injuries.

About the numbers

No official count sorts every disease this way. These are the best available estimates from public-health and rare-disease sources.

08

Gene-based treatments: how many people so far

Choosing drugs by genes is common already. Gene repair started with rare diseases and is reaching common ones.

Drug matched to genes

Targeted cancer drugs

Aimed at the tumor's own DNA change. 13.6% of US cancer patients were eligible for one by 2020, up from 5.1% in 2006.

Rewrites the DNA letter itself. 500+ patients on the first approved treatment for inherited blood disorders. Next focus includes heart disease: one infusion about halved “bad” cholesterol in early trials.

Gene editing in action

A guided tool finds the faulty letter, cuts it out, and the correct one goes in. Early trials can switch off a cholesterol gene with one infusion, aiming to prevent heart attacks.

09

Newborn screening, prenatal testing, tumor DNA assays and pharmacogenetics are delivered through doctors, hospitals and public-health programs — not consumer ads.

11

Why hospitals alone are not enough

Hospitals are built to treat illness. Understanding and steering your health also depends on life outside them.

≤5 minper doctor visit for half the world's people全球一半人口每次看医生的时间
80%of what decides your health happens outside the hospital (habits, living conditions, environment — estimate from a widely used US model)
14%of family doctors in a US survey felt confident reading a genetic test

Bring it to your doctor

A short visit goes further when you arrive prepared.

Adds to care — does not replace it补充医疗,而非替代

All of this adds to medical care and does not replace it: for symptoms, emergencies and treatment, see a doctor.

12

Where to look for technology you can trust

Reliable genetic information needs precise, proven technology. Here is what exists, and how to judge who offers it.

Before pregnancy · Carrier screening

Do both partners carry the same hidden variant?

During pregnancy · Prenatal testing from blood

The baby's DNA, read from the mother's blood.

At birth · Newborn genetic screening

Finds treatable conditions in the first days.

Adult life · Genome and drug-gene tests

One genome, re-read as new questions arise.

Earlier detection and tracking of treatment.

The precision benchmark

Prenatal testing must find the baby's DNA — about 1 piece in 10 of the DNA in the mother's blood — and still be right.

Where to check the answers

Regulators' lists of approved tests · Guidelines from medical societies · Published, peer-reviewed studies · Hospital genetics or prenatal diagnosis departments.

Five questions to ask any provider

  1. 1Is it registered or accredited? (e.g. NMPA in China, or CLIA and CAP in the US)有没有注册或认证?(例如中国的NMPA注册,美国的CLIA和CAP认证)
  2. 2Is there published proof? Peer-reviewed studies, and in how many patients.
  3. 3Is the method named, with its limits? What it can find, and what it can miss.
  4. 4Who explains the result? Counseling before and after the test.
  5. 5Where does your DNA data go? How it is stored, shared and deleted.
13

Frontier example: prenatal testing from blood

Non-invasive prenatal testing (NIPT) screens the baby from the mother's blood. Few genetic tests demand more precision.

~1 in 10DNA pieces in her blood come from the baby — often fewer的血中DNA片段来自胎儿,往往还更少
~5%is the whole signal an extra chromosome adds to the count
Millionsof DNA pieces are read and counted for every sample个DNA片段:每份样本都要读取并计数

Why it is one of the hardest tests

The result guides a pregnancy, so errors must be rare. Labs must separate a small fetal signal from a much larger maternal background.

Two ways to read it两种读取方法

Count everything: sequence random pieces from every chromosome and count them. Targeted sequencing: read thousands of chosen spots very deeply, using a fraction of the data. That depth helps separate the baby's DNA from the mother's and can reach smaller changes, even single genes.

How labs do targeted sequencing (educational note)

Clinical labs that offer targeted NIPT typically: (1) pull cell-free DNA from a maternal blood draw; (2) enrich or amplify selected chromosome regions or SNP sites; (3) sequence those sites at high depth; (4) statistically compare fetal vs maternal fractions and flag aneuploidy or selected microdeletion / single-gene signals. Accreditation (for example NMPA registration in China, or CLIA/CAP in the US), published validation cohorts, and clear reporting of test limits matter more than any brand label. This page names no commercial products.

Screening, not diagnosis

NIPT is a screening test, not a diagnosis: a high-risk result must be confirmed by a diagnostic test before any decision.NIPT

14

Start early, and study the family together

Inherited DNA is shared within a family and stays the same for life, so an early family record keeps paying back.

92%of conditions found by newborn genome screening are not on the standard newborn panel
10–20%of people over 70 carry blood-cell DNA changes gained in life; a baseline shows them的70岁以上人群,血细胞带有后天获得的DNA变化;有了基线,才看得出变化
2×the odds of finding the cause of a child's genetic disease when both parents are read too的几率找到孩子遗传病的病因:前提是父母也一起检测
~70DNA changes in each child are new — seen only by comparing child and both parents个DNA变化是每个孩子新产生的,父母都没有,只有三人对比才能发现
50% / 25%chance of sharing a carrier's high-risk variant: each child or sibling, then each grandchild

Start at the beginning of life从生命之初开始

Study three generations together. Grandparents' later-life illnesses reveal family risks decades ahead.

Read once

Inherited DNA does not change, so the first reading serves every later decision.

Act together as a family

Identify risk before symptoms, monitor changes against a baseline, and plan together. For children, guidance generally favors testing for conditions that can be acted on in childhood; other results can wait until they can choose.

15

For families who can afford it: why start now有条件的家庭:为什么现在就开始

The cost is now small next to what is at stake, and the value grows with time and with every relative tested.

$1,500–5,000buys a clinical-grade genome with a specialist's report (typical private-pay range)
1 in 75people carry a high-risk gene for cancer or heart disease; 90% of them had not been told
91% vs 13%five-year survival for colon cancer found early versus late — known risk means earlier screening
99.5%carry a variant that may change how a drug works — one test serves every future prescription
50%chance that each child, brother or sister shares a parent's high-risk variant

Why some families go first为什么有条件的家庭先行一步

For healthy people, genome testing is usually paid for privately, and so is the follow-up: extra screening, specialist advice, family planning.

Be realistic

Most results are reassuring, not dramatic, and a clear result does not rule out disease. Use accredited labs and genetic counseling, and check privacy and insurance rules.
